چکیده

Abstract Study question Do gut microbial composition and functionality differ between women with without endometriosis? Summary answer Gut microbiome diversity (species pathways) were not significantly different endometriosis. What is known already Endometriosis, defined as the presence of endometrial-like tissue outside uterus, one most common female reproductive disorders. Although theories possible causes endometriosis have been proposed, its pathogenesis clear. Novel studies indicate that may be involved in etiology endometriosis, nevertheless, connection microbes, dysbiosis development remains unexplored. This study aims to analyze compare profile a large cohort identify targets potentially disease. design, size, duration case-control included subsample 1000 (age = 45.61±10.36 years; BMI 25.67±5.59) Estonian Microbiome (EstMB) cohort, volunteer-based sub-cohort Biobank created 2017. 136 864 control who diagnosed or any prevalent comorbidities (systemic lupus erythematosus, rheumatoid arthritis, autoimmune thyroiditis, celiac disease, multiple sclerosis irritable bowel syndrome) this study. Participants/materials, setting, methods Microbial DNA from fecal samples was extracted sequenced by paired-end metagenomic shotgun sequencing (Illumina Novaseq 6000 platform). functional pathways annotated using Kyoto Encyclopedia Genes Genomes (KEGG) database (https://www.genome.jp/kegg/). Partitioning around medoids (PAM) algorithm performed cluster population. The alpha- beta-diversity differential abundance analyses assess KEGG orthologies [KO]) both groups. Main results role chance After metagenomics analysis, 17180 microbes 7869 KO detected. Those bacteria relative > 1 % used for analyses, resulting 2442 species 1974 KO. PAM clustering analysis stratified population into two enterotypes: characterized high Prevotella copri second presented Bacteroides genus (PERMANOVA, p 0.001). However, enterotypes associated presence/absence alpha-diversity (observed richness Shannon’s index) (all p-value 0.05). Beta-diversity on profile) indicated no significant dissimilarity groups all No nor detected after testing adjustment FDR Limitations, reasons caution did distinct A deeper considering potential confounders (specifically hormonal treatment patients) needed further confirm our results. Wider implications findings Endometriosis widespread disorder affecting ∼10% reproductive-age women. To best knowledge, biggest metagenome Our do find enough evidence support existence microbiome-dependent mechanism implicated Trial registration number Not applicable

Polycystic ovary syndrome (PCOS) is a complex endocrine disorder that affects 8-13% of women at reproductive age worldwide. This disorder is usually associated with menstrual disorders, infertility, obesity, and insulin resistance. The underlying cause of this syndrome is unknown, however in recent years, researchers have shown an association between intestinal microbiota alterations )dysbiosi...

Changes of the Intestinal Microbiome–Host Homeostasis in HIV-Infected Individuals – A Focus on the Bacterial Gut Microbiome

Human immunodeficiency virus (HIV) infections cause severe CD4+ T cell depletion leading to chronic inflammation and immune activation, impaired barrier function, and microbial translocation. Even under effective antiretroviral therapy, these processes persist, leading to gut microbiome dysbiosis and disturbance of microbiome-host homeostasis. This systematic review aims at analyzing how gut mi...

Protocol for the Emory University African American Vaginal, Oral, and Gut Microbiome in Pregnancy Cohort Study

Background: Adverse birth and neonatal outcomes disproportionately affect African American women and infants compared to those of other races/ethnicities. While significant research has sought to identify underlying factors contributing to these disparities, current understanding remains limited, constraining prevention, early diagnosis, and treatment. With the development of next generation se...

The gut mycobiome of the Human Microbiome Project healthy cohort

BACKGROUND Most studies describing the human gut microbiome in healthy and diseased states have emphasized the bacterial component, but the fungal microbiome (i.e., the mycobiome) is beginning to gain recognition as a fundamental part of our microbiome. To date, human gut mycobiome studies have primarily been disease centric or in small cohorts of healthy individuals. To contribute to existing ...
